MiR-141 inhibits colorectal cancer cell proliferation and invasion by targeting PRKAR1A

چکیده
Dysregulation of microRNAs (miRNAs) has been associated with the initiation and progression of oncogenesis in humans. Growing evidence has indicated that abnormal expression of microRNA-141 was associated with tumorigenesis and carcinoma progression of various cancers. In the present study, the miR-141 expression was assessed in CRC cell lines and the normal colonic cell using RTq-PCR, which demonstrated that miR-141 was significantly downregulated in CRC cells. Functional study revealed that overexpression of miR-141 significantly inhibited the proliferation, invasion and migration of CRC cells. Luciferase reporter assay validated that protein kinase A regulatory subunit 1 alpha (PRKAR1A) was a direct target of miR-141, Overexpression of miR-141 suppressed PRKAR1A protein expression, and rescued PRKAR1A attenuated the function of miR-141 in CRC cells. Taken together, our results highlight the significance of miR-141/PRKAR1A interaction in the development and progression of CRC cells, and suggest that miR-141 functions as a tumor suppressor in CRC cells. Targeting PRKAR1A may be an important mechanism involved.

miR-506 inhibits cell proliferation and invasion by targeting TET family in colorectal cancer
Objective(s): Ten-eleven translocation (TET) family members have been shown to be involved in the development of many tumors. However, the biological role of the TET family and its mechanism of action in colorectal carcinogenesis and progression remain poorly understood. Materials and Methods:We measured the expression levels of TET family members in colorectal cancer (CRC) specimens, in the c...
متن کاملMiR-27b directly targets Rab3D to inhibit the malignant phenotype in colorectal cancer
MiRNAs, as oncogenes or as anti-oncogenes, play critically regulated roles in the initiation and progression of colorectal cancer at posttranscriptional level. However, the underlying functions of miR-27b in colorectal cancer remain largely unexplored. Here, we demonstrated miR-27b is significantly down-regulated in colorectal cancer tissues, and decreased miR-27b expression was closely associa...
متن کاملThe Effects of NDRG2 Overexpression on Cell Proliferation and Invasiveness of SW48 Colorectal Cancer Cell Line
Background: Colorectal cancer (CRC) is one of the most common causes of cancer-related death in the world. The expression of N-myc downstream-regulated gene 2 (NDRG2) is down-regulated in CRC. The aim of this study was to investigate the effect of NDRG2 overexpression on cell proliferation and invasive potential of SW48 cells. Methods: SW48 cells were transfected with a plasmid overexpressing ND...

Objective(s): To explore the correlation between expression patterns and functions of miR-383 and LDHA in hepatocellular cancer (HCC). Materials and Methods: We detected the expression of miR-383 and LDHA in 30 HCC tissues and their matched adjacent normal tissues using qRT-PCR. Then we performed MTT assay, foci formation assay, transwell migration assay, glucose uptake assay and lactate produc...
متن کاملmiR-92a promotes hepatocellular carcinoma cells proliferation and invasion by FOXA2 targeting
Objective(s): MicroRNAs (miRNAs) are considered as powerful, post-transcriptional regulators of gene expression in hepatocellular carcinoma cells (HCC). However, the function of miR-92a is still unclear in HCC. Materials and Methods: Expression of miR-92a in human HCC cell lines was evaluated using qRT-PCR. MTT assay and transwell assay were used to examine the function of miR-92a in HepG2 and ...
